The Ni-Fe hydrogenase we use is an enzyme that can catalyze not only the reduction of hydrogen ions to hydrogen under anaerobic conditions but also the reversed reaction under aerobic conditions. Among this enzyme, hoxF is a hydrogenase subunit responsible for electron transport, which, together with the hoxU subunit, forms the module of NADH dehydrogenase.

And then it will eventually make up parts [https://parts.igem.org/Part:BBa_K5115066 BBa_K5115066(mineral, U module)]and [https://parts.igem.org/Part:BBa_K5115067 BBa_K5115067(mineral, F module)], which are used for further experiment.

The most important group in hoxF is FMN-b, which has the ability of switching electron. Under anaerobic conditions, NADH is oxidized to NAD<sup>+</sup> on the surface of hoxF subunit. Under aerobic conditions, NAD<sup>+</sup> is reduced to NADH on the surface of the hoxF subunit. Under anaerobic conditions, the electrons generated in this reaction travel through a series of processes to the hoxH, completing the final reduction of the hydrogen ion.

Introduction

The hoxF is a subunit of the hox and hyp operon BBa_K5115020.

Usage and Biology

It's used to build up the hydrogenase which reduces the nickel ion. In our experiment, it was constructed into our ribozyme-assisted polycistronic co-expression system BBa_K5115011(ribozyme+RBS+hoxF+stem-loop). And then it will eventually make up parts BBa_K5115066(mineral, U module)and BBa_K5115067(mineral, F module), which are used for further experiment.
