This part was designed to generate the cheZ proteins which are responsible for the dephosphorylation of cheY protein involved in bacteria flagella movement. A 6-histidine tail is added to the cheZ encoding region. The pTet can be repressed by the TetR repressor protein(C0040). It is constitutively on without the presence of TetR repressor. The backbone of this biobrick is based on BBa_J61003. CheZ_his tag and strong double terminators lie between XbaI and SpeI.

Thus, the expression of ''cheZ'' can be easily detected by immunostainning and western blotting analysis. pTet-rbs-chez_histag-double terminator


This part is an improved version of BBa_K094102. BBa_K094102 was designed to express CheZ protein, which is responsible for the dephosphorylation of CheY, a major effector in bacterial flagella rotation. cheZ gene is driven by the pTet promoter, whose expression is induced by adding anhydrotetracycline (aTc). The backbone of this biobrick is BBa_J61003. A strong double terminators lie between XbaI and SpeI sites.
