Design Notes
The 547th codon coding for S amino acid is deleted in order to diminish its sensitivity to leucine while maintainning its funciton since the presence of leucine can result in a decrease in the activity of LEU4. The composite part appends the first 26 amino acids of COX4 to the N-terminus of LEU4^S547∆ in order to relocate and overexpress LEU4^S547∆ in mitochondria.
