The design of HEK3 pegRNA includes the U6 promoter, spacer, guide RNA scaffold, rttpbs and polyA tail.
Source
The original HEK3 pegRNA design containing spacer and rttpbs sequence was reported in Prime Editor 2 paper cited in References. With the help of TWIST Bioscience we synthesized those and other necessary sequences, and further combined with 65777 backbone in our lab.
