BBa_C0051 DNA was used as themplate to site directed mutagenesis. Our aim was changing T450->C to remove premature stop codon and introducion of cIts857 mutation (TTTTGCAAGCAA -> TTTTGtAAGCAA) to make this variant of cI thermosensitive.
