This part is a time delay device using the regulatory promoter and the interaction between the inducer, repressor and regulatory promoter. The promoter pRE could be induced by BBa_K116602,the inducer CII, which results in the production of the TetR and green fluorescent protein. PenI from Bacillus licheniformis with LVA tail could repress the regulatory promoter BBa_R0074 pPenI, which results in the lower production of the inducer CII.

CII/PenI Damped Oscillator With GFP Reporter (II)

This device is produces damped oscillations between PenI and CII. The pRE can be activated by CII, which in turn produces PenI and GFP. The PenI then represses the pPenI resulting in slower production of CII.

This design was based on the design of a relaxation oscillator (two component oscillator). This design does not produce sustained oscillations like a normal oscillator (relaxation based or otherwise) because of the two oscillating effectors not being able to directly affect each other.


Modelling

The modeling graph would look like the same as the CII/TetR damped oscillator With GFP Reporter (I) with the only difference being that the rate of damping would occur faster if the (strength of pPenI)/(strength of CII) is smaller.
