This composite part is used for testing the presence of hsa-miR-145, hsa-miR-199a and hsa-miR-125b. It has a T7 promoter for expression of functional RNA BBa_K1614000. This is followed by the code for our AND-gate toehold switch BBa_K4642005 and the anti-miRNAs BBa_K4642006 and BBa_K4642007. Placing a reporter protein downstream of this toehold switch part will mean that when our trigger RNAs are present in high enough quantities, the protein in the CDS will be translated and the reporter protein can be observed. Our chosen reporter protein is mCherry BBa_J06504. This fluorescence produced can be detected with a luminometer to affirm a positive test. | This composite part is used for testing the presence of hsa-miR-145, hsa-miR-199a and hsa-miR-125b.
