For detailed design information, you can look up the registry of (BBa_K 4606003), the device for gene segment of SMAD4. According to the base sequence information of SMAD4 in the database, we mutated the SMAD4 gene in a region that complementary binds to miR-3074-5p. By inserting its 3’UTR region (414bp) behind the luciferase gene in the vector, we could determine the expression level of this gene segment.
