Tdc is the gene encoding tryptophan decarboxylase (TDC), which can remove the carboxyl group from 5-hydroxytryptophan (5-HTP) and convert it into serotonin (5-hydroxytryptophan, 5-HT) molecules. Tryptophan decarboxylase exists widely in animals and plants, especially in the central nervous system and gastrointestinal tract. Among them, 5-HTP is a precursor substance of neurotransmitter, while serotonin is an important neurotransmitter, similar to intestinal motor in terms of intestinal physiological function, and plays an important role in alleviating chronic constipation.

Our project uses the introduction of Tdc sequences and inducing their expression to produce serotonin to test their efficacy in treating constipation. The original sequence of TDC was derived from the genome of the organism Oryza sativa Japonica Group (Japanese rice).

In our 5-HT producing circuit, TDC and TPH together catalyze the generation of 5-HT, when sensing a decrease in butyrate concentration. Experimental results about our 5-HT producing circuit can be found in our composite part BBa K4656007.
