Tph1 is the gene encoding tryptophan hydroxylase (TPH), which plays an important role in the synthesis of serotonin (5-hydroxytryptamine, 5-HT), in which the rate-limiting step is catalyzed by TPH. The Tph1 gene expresses the TPH1 protein, which converts tryptophan (an essential amino acid) to 5-hydroxytryptophan (5-HTP).

As a neurotransmitter, serotonin can play the role of intestinal motility. If its secretion is insufficient, it can easily lead to the decline of intestinal motility, and then lead to chronic constipation. Therefore, constructing plasmids introducing Tph1 gene and promoting TPH expression is conducive to the treatment of chronic constipation.

TPH

TPH also plays a crucial role in the synthesis of serotonin, where the rate-limiting step is catalyzed by TPH to convert tryptophan (an essential amino acid) to 5-hydroxytryptophan (5-HTP).

The synthesis of 5-HT

Usage and Biology

As a neurotransmitter, serotonin plays a role in intestinal motility. If secretion in the gut is insufficient, it can easily lead to the decline of intestinal motility, and then lead to chronic constipation. Therefore, promoting TPH expression is conducive to the treatment of chronic constipation. Experimental results about our 5-HT producing circuit can be found in our composite part BBa K4656007.
