<p>pDawn(cI-LVA)-RBS070-X174E-T1 was inserted into <i>G. hansenii</i>  ATCC53582 by electroporation and identified. As shown in Figure 4, pDawn(cI-LVA)-RBS070-X174E-T1 was successfully inserted into <i>G. hansenii</i> ATCC53582.</p>

Usage

The pDawn (cI-LVA)(BBa_K1075044) blue light responsive system and the lysis gene X174E(BBa_K1835500) were inserted into the pSEVA331 expression vector, which was incorporated into the E.coli TOP10, screened for the bacterial colonies that grew in the dark but did not grow under blue light..

Characterization

1.Batch screening of pDawn-RBS070-X174E-T1 by evaluating the response to blue light.

As shown in Figure 2, the 11th and 15th bacterial colonies did not grow under blue light but grow in the dark. In order to further explore the lysis effect of pDawn(cI-LVA)-RBS070-X174E-T1, we tracked the OD600 value of pDawn(cI-LVA)-RBS070-X174E-T1 and plotted the growth curve diagram. As shown in Figure 3, the lysis effect of pDawn(cI-LVA)-RBS070-X174E-T1 is not well.
