Permeability device
Part Description
In this composite, the Lac promoter regulates the expression of phenylalanine permease which is a single integral membrane transporter that uses the proton motive force to antiport L-phenylalanine and L-tyrosine. And present naturally in Escherichia coli.
Usage
We implement this composite to increase the permeability of cells to phenylalanine by the expression of phenylalanine permease which is a single, integral membrane transporter to improve the sensitivity of our circuit to phenylalanine as shown in figure 1 and figure 2.
Figure 1. (shows the permeability circuit in our E.coli-based system.)
Figure 2. (shows the whole cell-based biosensor.)
Experimental Characterization
This figure shows an experimental characterization of this part as it's validated through gel electrophoresis as it is in lane 7. The run part (ordered from IDT) included Lac promoter - Permease.
