Lac promoter
Part Description
A CAP binding site and the lac operator sequence lacO are present in this lac promoter. Although the lac promoter is not as potent as the tac or trc promoters, it nevertheless provides for decent quantities of foreign protein production in high copy-number vectors. Expression from the lac promoter is activated in the absence of the lac repressor (LacI).
Usage
We use the lac promoter as a constitutive promoter for the regulation of (Phenylalanine-specific permease)expression even if it is not as powerful as the tac or trc promoters. It still shows significant levels of phenylalanine enough to induce and start our circuit activity by improving the permeability of the membrane to phenylalanine as shown in figure 1.
Experimental Characterization
This figure shows an experimental characterization of this part as it's validated through gel electrophoresis as it is in lane 7. The run part (ordered from IDT) included Lac promoter - Permease.
