The gene fragment Pre B1 is composed of Pre A1 and the TetR gene <partinfo>BBa_C0040</partinfo>. We digested the vector fragment Pre A1 with SpeI and PstI; the insert fragment <partinfo>BBa_C0040</partinfo> with XbaI and PstI. We ligated these fragments together to form Pre B1.
