We designed the sgRNA (19 bp) that can bind to the T7 promoter, which can guide the dCas9 to bind to the T7 promoter and inhibit the function of the promoter.
How does it work?
Compared with traditional Cas9, dCas9 does not have the function of knockout, but only retains the ability to be guided into the genome by sgRNA. dCas9 can fusion or recruit transcription factors.
