6xHis-Tag for Purification and Identification of Protein of Interest
The His-Tag is a commonly used sequence both as a reporter and as a purification tag. Its sequence, consisting of 6 histidine residues, has a very high affinity to nickel(II) ions and is therefore used in affinity purification columns with immobilized nickel ions. The binding of the His-Tag to the nickel ions is not efficient at acidic pH values, and any purification process should therefore be done above pH=7. Imidazole is used to elute the protein from the column by competing with the His tag on binding to the immobilized Nickel ions.
