5CON1(H)_NS3-up (PCC 11801)
This part is an homology 5’ connector harboring the up homology arm for the homologous recombination within the Neutral Site Nº 3 of PCC11801.
Within the Marburg Collection design, connectors are found in the upstream (5’) and downstream (3’) regions of the level 1 assemblies, flanking the transcriptional unit. This allows the further assembly of multiple transcriptional units into a level 2 genetic construct using level 1 parts flanked by different connectors. To know more about, read the design principles of Marburg Collection: https://2019.igem.org/Team:Marburg/Part_Collection.
Among other functions, connectors can also be used to include homology regions for integrative recombination in the chassis genome. This way 5’ Connector parts can harbor the up / down homology arm required for double homologous recombination.
The part has been designed in accordance with the Marburg Collection guidelines. 5’ / 3’ Connectors correspond with the parts found Upstream / Downstream of the level 1 assembled transcriptional unit.

The genomic integration loci targeted by this part corresponds with a novel neutral site for Synechococcus PCC11801. This neutral site has been identified by the MADRID_UCM 2021 iGEM team as a part of their project.
