In an attempt to increase the sensitivity of [https://parts.igem.org/Part:BBa_K3725130 BBa_K3725130] to extracellular phosphate, we designed part [https://parts.igem.org/Part:BBa_K3725150 BBa_K3725150], a phosphate-sensitive biosensor with modified promoter BBa_K3725140 and GFP reporter. The new promoter utilizes a potentially more efficient consensus sequence by directly mutating the RNA polymerase binding site. The mutation affects the construct’s ability to transcribe downstream GFP, thus increasing the sensitivity of the construct and potentially creating an enhanced characterization curve.

PhoB Redesigned Mutated Consensus Promoter

DESIGN

In an attempt to increase the sensitivity of BBa_K3725130 to extracellular phosphate, we designed part BBa_K3725150, a phosphate-sensitive biosensor with modified promoter BBa_K3725140 and GFP reporter. The new promoter utilizes a potentially more efficient consensus sequence by directly mutating the RNA polymerase binding site. The mutation affects the construct’s ability to transcribe downstream GFP, thus increasing the sensitivity of the construct and potentially creating an enhanced characterization curve.
