RNA Part of an RNA·DNA-DNA triple helix + BoxB
This part consists of a sequence that forms an RNA·DNA-DNA triple helix (DNA part: BBa_K3657023) and a sequence that forms a BoxB hairpin (BBa_K3657015).
The BoxB hairpin binds to the lambdaN protein (BBa_K3657014). The triple helix forming part binds the DNA sequence specifically. In that way this part should theoretically attract a protein of interest fused to a lambdaN protein to a specific region on DNA. Due to the COVID19 pandemic we were not able to get any empirical evidence that this part functions as described above. More information available here: iGEM Heidelberg 2020.
Figure 1: Secondary structure of BBa_K3657052
MS2 hairpin forms a stable structure. The triple helix forming sequence doesn't form any stable structure and does not interfere with the BoxB hairpin. The structure was generated by the Geneious software.
