Applications of BBa_K3365054

We add this part into the off-target detection platform(part BBa_K3365062, BBa_K3365063, BBa_K3365064 and BBa_K3365065) as a reporter by the following three steps:

Firstly, we add target sequence to the head of spacer by PCR: primers used in this procedure contains the target sequence. Secondly, we use Overlap PCR to connect target and eGFP fragment. The overlap PCR is used for fear that target-blank fragment is too short to be attached to the eGFP sequence. Thirdly, we obtain terminator by performing PCR on pE1a-RFP and then link target+blank+eGFP with terminator through overlap PCR.
