Forward primer with BBa_B0034 for amplifying BBa_E0040
A forward primer that contains RBS34 which is 60 base pairs long that can be used to amplify the GFP part BBa_E0040 by performing PCR. It can be used to characterise the RBS activity easily by measuring its fluorescence.
This will be used as a standard in measuring RBS activity as RBS strength is often measured relative to RBS34. This primer contains an XbaI restriction enzyme site, a 21-nucleotide random sequence, RBS34, another random sequence for binding, then the first 21 nucleotides of the GFP sequence
