CRISPR Array targeting the Plasmid DNA of the Exendin-4 expression system controlled by Lsr
The CRISPR array sequence needed to guide the Cas3 nuclease to its cut sites on the plasmid with the glucose dependent Exendin-4 expression system under the control of the LsrR promoter. When LsrR is present the array is not transcribed and the CRISPR/Cas3 System lacks its gRNA and is inactive. Combined with the fatty acid responsive repression system (BBa_K3096046) the expression of the array can be controlled.
