Design Notes
To reduce sensor basal background, we inserted an extra NsrR binding sequence (NsrRBS) downstream of PyeaR to create a ‘roadblocking’ effect. The distance between the two NsrRs , the sensor’s leakiness and sensitivitcan be adjusted. However, due to time limitation we only modified 47 bp between two NsrRBSs.
