pMreB

MreB, a homologue of actin, is a protein which is expressed in nearly all rod-shaped bacterial cells. It is one of the three types of actin-like filaments present in bacteria. MreB is important for maintenance of cell width and cell viability as it recruits the machinery responsible for the establishment of peptidoglycan, an essential building block for the cell wall. iGEM Leiden 2018 has created a screening system for the detection of new antibiotics. We have tested several stress-activated promoters against a repertoire of known antibiotics (Figure 1) using flow cytometry. We fused promoters to green fluorescent protein and stressed transformed DH5α cells for several hours.

As can be observed in Figure 1 the MreB promoter is only slightly activated by nalidixic acid and tetracycline
