Peptidyl Prolyl Isomerase (PPiase) in pSB1C3
Peptidyl-prolyl isomerase (PPiase) is a protein that aids the isomerization of prolines in order to catalyze the addition proline amino acids onto the peptide chains from the aminoacyl-tRNA it is attached to [1]. This part has been codon optimized for use in E.coli , Contains a N-terminal his tag, and has a T7 promoter, medium strength RBS, and double terminator. This part was originally designed by the 2017 LEthbridge iGEm team (http://2017.igem.org/Team:Lethbridge).
This part was improved by placing it into the pSB1C3 plasmid.
Figure 1- An overexpression gel of PPiase protein run on a 12% SDS-PAGE. Samples were taken at point of induction and then in 1 hour increments thereafter. The induced proteins is expected to be seen at about 22kDa range
