We grew colonies on three different Petri dishes over night in LB amp media. Each Petri dish tested a different part of the experiment. One was a positive control (labeled P) that would produce red colonies. Another was the negative control (labeled N) that would grow green colonies. The last Petri dish (labeled X) would contain our transformed plasmids containing our mutated gene sequence. Then we harvested three different colonies from the X dish, a colony from the P dish, and a colony from the N dish. These were grown overnight in a LB amp media solution. We then took triplicate samples from each test tube and measured the fluorescence and cell density of each.
