The weak constitutive promoter J23116 will produce a steady stream of tetR, which (in the absence of aTc) repress transcription of the pTet-cis3-Q transcript. The riboregulator design further represses levels of Q protein in the cell. The transcript will form a cis-hairpin loop and prevent translation in the absence of a riboregulator trans sequence, which binds to and opens up the cis-hairpin, allowing the ribosome to access the RBS site.
