Promoter for Bacillus megaterium
Description
This part is a promoter used in Bacillus megaterium. We got its sequence in pSPAsp-hp, which is a plasmid used for protein expression in Bacillus megaterium. Then the whole part has been synthesized by AuGCT.
Verification
As for the verification of this promoter, we combined it with BBa_K2462001 and BBa_K2462000 in order to make a circuit. We planned to test this promoter through the test of our enzyme's effect. Several experiments have been done, and details can be found in the following two pages:
https://parts.igem.org/Part:BBa_K2462001
https://parts.igem.org/wiki/index.php?title=Part:BBa_K2462000
The results of experiments are depicted as following:

Disscusion
This year, our team decided to carry on a project using Bacillus megaterium as the host. Through our own experience, we felt that there's few parts in the past years related to the expression in Bacillus mageterium although it's a great host for many protein to be expressed. Thus, when making parts, we considered more about filling this blank and give the following teams convenience to use Bacillus megaterium. Unfortunately, we are not able to verify this part successfully, but we do welcome other teams to use our part and hope our parts can bring them help. As this promoter was from pSPAsp-hp——a plasmid in Addgene, few articles using this promoter can be found in their page for other teams’ reference.
