Usage and Biology

The sequence of this part is designed and offered by Jef D. Boeke’s laboratory. And we used the mailed plasmid to test its function. As we can see from the figure below, it is obvious that this modified promoter can  transcribe the CDS normally without the phlF in the Saccharomyces cerevisiaea.

Promoter(phlF regulated)

This part is an inducible promoter regulated by PhlF. And the sequence of it is offered by Jef D.Boeke's laboratory.

It is a modified ADH1 promoter from the yeast Saccharomyces cerevisiaea having a phlF operator sequence (phlO) embedded downstream the promoter. The phlO region can be bound by phlF, a regulatory protein existing in Pseudomonas fluorescens and related species.

In the absence of PhlF protein, this part promotes transcription.

In the presence of PhlF protein, this part inhibits transcription.

PhlF can be inhibited by 2,4-diacetylphloroglucinol (DAPG).

PhlF can be coded by BBa_K2207009.
