FepA L8 Mutant with Elowitz RBS. This is an intermediate on the way to construction of a permeability device for the outer membrane of E. coli.

This device employs the FepA L8T mutant described [https://parts.igem.org/wiki/index.php?title=Part:BBa_I746200 here] to allow molecules larger than normal to pass through the outer membrane of E. coli. The porins usually present will allow molecules up to around 700Da to freely diffuse in and out, however with this device, molecules of at least 1400Da should be able to pass through. Permeability was tested using the antibiotic Bacitracin. Wild type E. coli are resistant to this as the molecule is 1422Da in size. Expressing the device made the bacteria sensitive to bacitracin. In this device, the mutant FepA is under the control of a very high strength promotor.

This device employs the FepA L8T mutant described here to allow molecules larger than normal to pass through the outer membrane of E. coli.
