B0015:S03706
A strong (as compared to J23116) constitutive promoter J23100 produces a steady stream of the tet repressor protein. When cloned into other constructs containing pTet, this tetR will constantly repress transcription of the pTet-regulated gene. This intermediate also contains a B0015 terminator upstream of the tetR producing region, so that pTet-spacer-cro/N/Q/YFP constructs can be easily cloned upstream of the terminator, resulting in the final testing construct.
