sfGFP under proC promoter with a hairpin in 5'UTR

A hairpin introduced to 5’UTR of proC slightly lowers transcription/translation. We are planning to extend it to use it as a thermometer, as for now we do not see temperature dependence.

This is the potential hairpin structure.

Results

proC [BBa_K1741012]

proC with a hairpin in 5'UTR [BBa_K1741013]

proD [BBa_K1741014]

The sfGFP fluorescence [RFU] was measured using Tecan fluorometer.

We observed that proD is up to three times stronger than proC. The hairpin concept has to be further elaborated upon and improved so as to stop translation in lower temperatures.
