This part is designed to contain two basic short sequences which can bind TALE1 and TALE2(3) specifically. These two basic sequences are repeated with no space by ten times, so it can bind more TALE protein and the spatial location of each pair of TALE1 and TALE2(3) is almost opposite.

Design Notes

The length of DNA BMs and intervening sequences is designed based on the typical in vivo B-type DNA structure, and the Structural Basis for Sequence-Specific Recognition of DNA by TAL Effectors.
