When connection k1555000 and RFP, there was no capable restriction enzyme to use. So we need to add capable restriction enzyme on primer to solve it.

We use promoter activated by cueO first. But after discussion with NJAU, we found that cueO promoter was not very specific on copper ion. So we changed to use copA promoter. Which is more specific.

Design Notes
