RGE

It contains GFP codon optimised to work in E.coli synthesised as the reverse complement, followed by a ribosome binding site also reversed Part BBa_B0034 After the RBS is the 3’UTR SLD3 containing a ribozyme Part BBa_K1442114 which is an RDRP promoter and is the test subject used in this testing module. This can be exchanged to test other promoters using the AflII and BsiWI restriction sites either side.

Each 3' promoter has a hammerhead ribozyme following it in order to create a clean slice with no scar removing the T7 terminator following transcription.
