After further investigation, the addition of the .015% hydrogen peroxide to the samples including the SoxS promoter and the negative control appears to have killed all of the cells. The incubation period was approximately 21 hours. The concentration of hydrogen peroxide or the duration of the incubation period, therefore was too high in order to simply increase oxidative stress, and ultimately produced ineffective results. The data is inclusive whether the soxR protein successfully repressed the SoxS promoter, or the cell was simply incapacitated by the addition of the hydrogen peroxide, or by the length of the incubation period. The graphs of the data are above. The first graph includes the positive control, but the second graph excludes this sample in order to make the ratios of the experimental and negative control samples more visible. The error bars indicate the standard deviation between the ratios of the samples.
