DarR operator

This part is the binding sequence of the c-di-AMP-dependent regulatory protein DarR. Binding of DarR to the DarR operator prevents transcription of the downstream information.

The DarR operator is a 14 bp palindromic DNA sequence discovered in Mycobacterium smegmatis. In M. smegmatis, the DarR operator is found upstream of darR (auto-regulation) and other genes. The sequence motif of the DarR operator varies slightly dependent on the downstream gene (Zhang et al., 2013). To create this biobrick, the operator sequence found upstream of darR was used.


The important information: The operator is not in the pSB1C3 vector but instead in the pBluescript -KS plasmid. This plasmid provides an ampicillin resistance. Furthermore, it has got a pUC origin.
