RcnR is a protein that constantly represses PrcnA. When there is nickel in the media RcnR alters its form, making it impossible to bind PrcnA, activating the production of the HoxN protein. This last protein is an efficient nickel transporter from ''Cupriavidus metallidurans'' that places in the membrane allowing the metal to get into the cell.

Prcna, hoxN

Prcnr, rcnR and PrcnA genes were used from E.coli regulation system.

Prcnr is a constitutive promoter, controlling the production of RcnR protein which

represses PrcnA. When there is nickel in the medium RcnR this induces a conformational

change in RcnR which prevents its binding to PrcnA, activating the production of the HoxN

protein in our construct. HoxN an efficient nickel membrane transporter form Cupriavidus

metallidurans that imports the metal into the cell. RFP was transcriptionally fused to HoxN

to monitor its expression.

Therefore, our system continuously detecting for nickel, and when this metal is detected,

it is absorbed.
