Long reporter top strand with bulge MammoBlock
Long reporter top strand with a 4 nucleotide bulge, binds to Part:BBa K779502.
Modifications: 5' Iowa Black RQ quencher, 3' Alexa 488 florophore, all bases modified with 2'O Methyl (ordered as a DNA oligo with Us instead of Ts).
IDT DNA oligo order: /5IAbRQ/mCmAmCmCmCmAmCmUmCmCmCmAmCmUmUmCmUmCmCmCmAmCmCmA/3AlexF488N/
MIT iGEM 2012 is still collecting data on this part.
Usage and Biology
Used as an alternative quencher (top) strand to Part:BBa K779501. The only difference is a 4 nucleotide bulge, which is supposed to protect the annealed complex (below) from degradation by the RISC complex (see the Design section).
When annealed to Part:BBa K779502 forms a tagged reporter for strand displacement reactions (input Part:BBa_K779503, negative control Part:BBa_K779504).
