Luminesensor repressible RecA408 promoter+CheZ
CheZ is a significant protein in chemotaxis pathway of E.coli. It can prevent the cells from tumbling around and keep the cells moving. Deletion of CheZ or repression of CheZ expression can result in a non-motile phenotype.
CheZ placed under RecA408 promoter (see BBa_K819002 constructed by Peking 2012 iGEM) can be repressed by LexA408 protein or protein with LexA408 domain(e.g. optimized Luminesensor BBa_K819005 constructed by Peking 2012 iGEM team), excluding the influence of endogenous LexA, forming an orthogonal modular biobrick. Thus the motility of E.coli can be well controlled with natural light (with the optimized Luminesensor) or other sources of LexA408 protein
Usage and Biology
Need to be used in ΔCheZ strain to exclude the influence of endogenous CheZ
