bacteria Intra-strain communication: An E. Coli containing BBa_K553010 constitutively produces the OC12 AHL quorum sensing molecule (strain A). The ammount of this molecule is enough to induce the activation of part BBa_K553008 (production of OC8 AHL) in a co-cultivated bacteria (strain B).
The following assay demonstrates that the organic extract of a co-culture media contains a considerable ammount of OC8.
The diffusion from the center of the OC8 AHL generate, in fonction of its concentration, the activation of a biosensor (A. tumefacines) in Xgal LB agar dishes.
HPLC-MS/MS assay of an organic extract of strain A with BBa_K553010 in a medium copy number plasmid.
Blue: culture media 20x concentrated + OC8 and OC12 AHL Standard 1uM - 10 uL loaded
