This gene comes from the hok/sok system in Escherichia coli. It is able to encode a type of antisense RNA that can specifically bind to the hok mRNA, repressing its translation. Therefore, the toxic protein cannot be produced in normal conditions, and the engineered bacteria will survive. It's worth paying attention is that the half-life of sok RNA is much shorter than hok mRNA, which lead to the situation that when both of them cannot express, sok RNA is completely degraded while mok mRNA still exist, making the toxic protein be encoded and bacteria be killed. We take advantage of this characteristic to design a suicide system.
