This part consists of a fusion protein Lpp-rbs-protein under the pLac promoter. The Lpp is a signal peptide. The ompA domain of the fusion directs the protein to the surface membrane. The pLac promoter can be repressed by LacI protein. This part lacks a terminator.

To simplify the the procedure and directly observe the expressed protein, we inserted a eGFP-coding sequence lacking a stop codon immediately upstream of streptavidin region. lpp_ompA_eGFP_streptavidin


This part is an improved version of BBa_J36846. BBa_J36846 consists of a signal peptide of lipoprotein (lpp), five transmembrane domains of outer membrane protein A (ompA), and streptavidin, wild-type with His6 tag. It's designed to express streptavidin on the cell surface. To test if the protein successfully expressed on the outer membrane of bacteria, you might need to perform immunostainning analysis or cell fractionation and western blot.
