This device is an improved version of BBa_K094102. It was designed to express the CheZ, which is responsible for the dephosphorylation of CheY involved in bacterial flagella rotation. Since lacking an antibody to recognize the expressed CheZ, we fused a His6-tag to the ''cheZ'' encoding region. We put'' cheZ'' gene under the control of the pTet promoter, which is repressed by the TetR repressor protein(C0040) and can be induced by adding anhydrotetracycline (aTc). The backbone of this biobrick is BBa_J61003. The strong double terminators lie between ''Xba''I and ''Spe''I sites.

BBa_K094102 was designed to express CheZ protein, which is responsible for the dephosphorylation of CheY, a major effector in bacterial flagella rotation. cheZ gene is driven by the pTet promoter, whose expression is induced by adding anhydrotetracycline (aTc). The backbone of this biobrick is BBa_J61003. A strong double terminators lie between XbaI and SpeI sites.

Thus, the expression of cheZ can be easily detected by immunostainning and western blotting analysis.
