Photorhabdus virulence cassette (PVC) is one group of extracellular Contractile injection systems (CISs) that are cell-puncturing nanodevices for targeted delivery of proteins .[1]Structurally similar to other CISs, PVCs are also macromolecular complexes containing a rigid tube structure housed in a contractile sheath, which is anchored to a baseplate and sharpened by a spike protein.[2](Figure1)

Pvc16 is a tube terminator,which possesses a central core and six arms.Six helices from the Pvc16 hexamer completely seal the central channel of the tube, leaving a narrow pore with a diameter of 7 A.Each Pvc16 monomer exhibits a dumbbell-like structure. Each Pvc16-NTD interacts with three copies of Pvc1.Pvc16-CTD stabilizes the sheath-tube complex at the capping end.The N-terminal domain (NTD) and C-terminal domain (CTD) is connected by a middle linker (MD).
Significantly,the binding of Pvc16 onto the PVC complex might not require the help of outer sheath and the capping of Pvc16 on the distal Pvc1 ring might set the signal to initiate the sheath loading.
After the tube reaches optimal length (likely controlled by the tape measure protein Pvc14), Pvc16 interacts with the topmost inner tube layer to terminate the tube elongation.
