piG_10b (tetR_bluB_T7RBS_riboK12_mazF)
Plasmid piG_10 is made up of the tet promoter/repressor, bluB, a riboswitch for AdoCbl, the tet RBS, mazF (toxin) and the rrnB terminator. The backbone we utilized was pGGAselect, a plasmid commonly used with golden gate cloning that has a chloramphenicol resistance. This BioBrick is BBa_K4604021 with an additional RBS.
Notice
We have performed western blot after induction of this BioBrick, however we could not detect any BluB expression. The reason for this is not known to us, however for the sake of completion and to allow further exploration, we wanted to upload this part.
