piG_10a (tetR_bluB_riboK12_mazF)

BioBrick piG_10a consists of the tet promoter/repressor, the modified T7 RBS, bluB, a riboswitch for AdoCbl, mazF and the rrnB terminator. The backbone we used in the experiments is pGGAselect. The tet promoter is a BioBrick from iGEM Freiburg 2022 (BBa_K4229059).

Notice

This BioBrick is the closest we got to testing all the parts our system consists of combined. Read more about the full setup on Ba_K4604023. However, performed western blot after induction of this BioBrick showed only BluB expression, no MazF. The reason for this is not known to us, however for the sake of completion and to allow further exploration, we wanted to upload this part.
