Design Notes

The design of RNF2 pegRNA included the following considerations. CACC overhang before the spacer, rttPBS length, the A to G edit, and polyA tail.

Source

The original RNF2 pegRNA sequence containing spacer and rttpbs was reported in Prime Editor 2 paper cited in References. We used those sequences and with the help of and TWIST Bioscience synthesized the parts, which were further pieced together with 65777 backbone in our lab.
