MaSp2 encodes for spider silk protein, which is a highly repetitive amino acid sequence and has the properties of being strong, flexible, and elastic. This amino acid sequence is relatively short compared to other amino acid sequences for spider silk proteins listed in the paper by Tian et al, 2003. The properties of MaSp2 in our project are to make the scoby membrane more flexible and softer. Our team converted this amino acid sequence into DNA sequences and consulted with the Mission Biotech company where we ordered the MaSp1(BBa_K4650002) and MaSp2 from. The company suggested our team to use several different 3-nucleotide codons encoded for one amino acid in these repeated amino acid sequences, so we would not get the same repeated DNA sequence. After consultation by the company, the team has made those MaSp1, and MaSp2 synthetic DNA along with the functional primer sets for MaSp1 and MaSp2 PCR technique. Our team also added the start codon, ATG, at the 5'end of the MaSp2 DNA sequence so the RNA transcription would start from ATG on MaSp2 DNA.
