The Crte-cytoTDS-MBP construct contains several genes. CrtE is the gene which encodes geranylgeranyl diphosphate synthase, an essential enzyme that synthesizes GGPP.  Taxadiene synthase is the enzyme which makes the precursor to paclitaxel, taxadiene, from GGPP.  The design of the Maltose binding protein coding sequence attached to the 3’ end of the TDS coding region is intended to assist with protein solubility in the cytosol. immediately upstream of the the Crte-cytoTDS-MBP construct is a Gmubi promoter, and downstream of the construct is an AtHSP terminator. The RUBY marker, which is used to visualize gene transformation, is also flanked by Gmubi and AtHSP respectively.
